Is this simply a tuning or are there mechanical differences?
My concern is the added work load on the tuned engine over time.

Yes, the difference between the 340 and 380 PS versions of the AJ126 is entirely the tune, regardless of which JLR model the engine is used in.
Even the "tuned" 380 PS version is under-stressed so no added workload to worry about.
Also remember that for a couple of years (2018-2019 IIRC) there was a factory 400 PS version available in the F-Type known as the 400 Sport.
The engine in my F-Type started off as the 380 PS version (ie F-Type V6S) which I then tuned to 412 hp with a "tune only" then a year after that I added a larger crank pulley and tune to take it to 450 hp. That was four years ago now and it has never missed a beat and has given me zero problems.
The same tuner (Velocity AP) now has a 470 hp pulleys and tune package for sale, the extra 20 hp coming from a new improved and smaller SC pulley and associated tune on top of the larger crank pulley.
And it's not really about the maximum hp, the big change is the massive increase in torque especially "under the curve" which improves mid-range response and drivability no end.
Last but not least no JLR dealership will ever tune your 340 PS version to 380 PS (or more), in theory they could but in practice they are forbidden to do so and are prevented from doing so. There are some privateers out there with the SDD equipment and knowledge who in theory could apply the 380 PS tune to older models but I'm not aware of any having the factory 380 PS tune to do this.
