First, unless you're buying the pre-mixed stuff, it has to be mixed 50/50 with water. And you should use distilled water. Tap water has stuff in it that can contribute to corrosion within the engine and radiator.

There's no telling how much fluid was still stuck in various chambers in the engine, hoses and radiator. The best way to do it would be to just fill it up to the fill line, run the engine for 30 seconds or so to let it circulate and "burp" all of the air, then fill it up to the fill line again. Repeat until the fluid level doesn't drop after running it for a minute or two. Don't let it get too hot, though.

Oh yeah... if you overfill it it will just vent the excess fluid into a canister that is located in the right front fender behind the wheel well liner. And then that canister has an overflow tube that will vent the fluid out onto the ground if it gets too full. I don't think there's much harm in overfilling it. That's much better than not having enough.

But maybe I'm wrong about that. I'll defer to one of the experts on that issue. Personally I wouldn't worry about too much fluid, but I'm a slacker.

I would let it run till it gets to operating temp, that way you know the t-stat opened and coolant is flowing through the engine properly.

Also, do not leave the system unpressurized (cap off), as this might leave air pockets in the system.
